The power of metabarcoding: Can we improve bioassessment and biodiversity surveys of stream macroinvertebrate communities?

Most stream bioassessment and biodiversity surveys are currently based on morphological identification of communities. However, DNA metabarcoding is emerging as a fast cost-effective alternative for species identification. We compared both methods in survey benthic macroinvertebrate communities across 36 sites northern Finland. identified 291 taxa which 62% were only by metabarcoding. produced extensive level inventories groups (Oligochaeta, Chironomidae, Simuliidae, Limoniidae Limnephilidae), was not feasible due to the high expertise needed. Metabarcoding also provided more insightful taxonomic information occurrence three red-listed vulnerable or data deficient species, discovery two likely cryptic potentially new Finland insect genera at an early larval stage that could be separated morphologically. it systematically failed reliably detect gastropods easily The impact mining community structure shown using suggests finer detail can improve detection subtle impacts. Both generally exhibited similar strength community-environment relationships, but showed better performance with presence/absence than relative sequence abundances. Our results suggest holds promise future anthropogenic assessments, although, our case, did much from key advantage lies efficient surveys, taxonomical studies applications conservation biology.
